Correlating Raman and fluorescence lifetime imaging

 

In collaboration with Becker & Hickl GmbH, we will present combined Raman spectroscopy and fluorescence lifetime imaging (FLIM) on a Renishaw inVia™ confocal Raman microscope.

FLIM allows the molecular environment of biological cells and tissue to be studied. Changes in pH, ion concentration and molecular binding can be detected. FLIM is a fast technique, so it provides an effective precursor to highly specific chemical analysis via Raman imaging. FLIM is compatible with Raman spectroscopy as it utilises autofluorescence, so it can be used for label-free sample analysis.

Within this webinar, we will describe how we have integrated FLIM hardware, developed by Becker & Hickl, onto a Renishaw inVia Raman microscope. Both techniques employ the same optical arrangement and mapping stage hardware. This means that FLIM and Raman data can be collected from the same sample area with pixel-to-pixel accuracy.

We will explain how correlative FLIM/Raman can analyse different materials and can be used for initial identification of areas with interesting molecular interactions. Then, we explain how targeted Raman spectroscopy is used to gain an understanding of the underlying biochemistry.

Overall, we will demonstrate a solution for performing both Raman and FLIM on a single microscope system. This enables detailed physical and chemical information to be collected from samples with ease.
